A morphological study on the cytotoxic effect of nicotine on the development of mouse early embryo in vitro





Abstract
This study was performed to examine the cytotoxic effect of nicotine on the various developmental stages of mouse early embryo. Two-cell embryos were cultured for 72 hours in the media containing various concentrations of nicotine.
The results obtained were as follows
1. Two-cell embryos were completely inhibited the differentiation to blastocysts at 5 X 10¢¥ M nicotine concentration when two-cell embryos were cultured for 72 hours in the media containing various concentrations of nicotine.
2. The longer the embryos exposed to nicotine, the more damages appeared in mouse early embryos treated with nicotine.
3. Morphological changes such as the fusion and fragmentation of blastomeres, cytolysis and developmental retardation in embryonal cells damaged to nicotine were more increased than those of the control group dependent on time and dose.
From these results, it suggests that nicotine represents cytotoxic effect on the various developmental stages of the mouse early embryos.
